Lyophilised semaglutide is generally held at -20 °C or below, protected from light and moisture. Reconstituted solutions are typically kept at 2-8 °C and used within a defined window because degradation accumulates over time. Repeated freeze-thaw cycles are discouraged, since each cycle can promote aggregation and reduce monomeric content. Room-temperature stability of the solid has been examined in some studies but remains incompletely characterised for long durations, so cold storage is the conservative default for research material.
Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.

Development began in the early 2010s with the goal of extending GLP-1 activity beyond the brief window achieved by native peptide infusion. The earliest approved formulation was a subcutaneous injection given once weekly. A later oral tablet pairs the peptide with an absorption enhancer, sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, usually shortened to SNAC. That carrier lowers local pH and helps the peptide cross gastric tissue. Both routes deliver the same active molecule.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, a gut hormone released after meals. Its backbone retains the GLP-1 sequence but incorporates two substitutions that slow enzymatic breakdown by dipeptidyl peptidase-4. A short polyethylene glycol linker and a C18 fatty diacid are attached to the peptide chain, allowing the molecule to bind serum albumin and remain in circulation far longer than the native hormone. The result is a circulating half-life measured in days rather than the minutes typical of endogenous GLP-1.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Lyophilized peptide material is typically stored at or below -20 °C, with -80 °C used for longer-term archives. Vials should remain sealed and desiccated because moisture promotes aggregation and hydrolysis. Repeated freeze-thaw cycles are avoided since they can alter peptide conformation and reduce recovery. Once reconstituted, solutions are generally kept at 2-8 °C and used within a defined window. Stability beyond those windows depends on buffer composition and concentration, and exact limits are product-specific rather than universal.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
== Genetics == Multiple epiphyseal dysplasia (MED) encompasses a spectrum of skeletal disorders, in which are inherited in an autosomal dominant form. However, there is an autosomal recessive form. Associated genes include COL9A1, COL9A2, COL9A3, COMP, and MATN3. Types include:
Most animals rely on biomass and bioenergy produced by plants and phytoplanktons (collectively called producers) through photosynthesis. Herbivores, as primary consumers, eat the plant material directly to digest and absorb the nutrients, while carnivores and other animals on higher trophic levels indirectly acquire the nutrients by eating the herbivores or other animals that have eaten the herbivores. Animals oxidise carbohydrates, lipids, proteins and other biomolecules in cellular respiration, which allows the animal to grow and to sustain basal metabolism and fuel other biological processes such as locomotion. Of the 550 gigatons of carbon biomass on Earth, animals account for 2 gigatons. Some benthic animals living close to hydrothermal vents and cold seeps on the dark sea floor consume organic matter produced through chemosynthesis (via oxidising inorganic compounds such as hydrogen sulfide) by archaea and bacteria. Animals originated in the ocean; all extant animal phyla, except for Micrognathozoa and Onychophora, feature at least some marine species. However, several lineages of arthropods begun to colonise land around the same time as land plants, between 510 and 471 million years ago, during the Late Cambrian or Early Ordovician periods. Vertebrates such as the lobe-finned fish Tiktaalik started to move on to land in the late Devonian, about 375 million years ago.
Lutetium (177Lu), which decays with the emission of a low-energy electron, is useful for treating some cancerous tumors. Several compounds of 177Lu are available for this purpose: 1) Lutetium chloride, LuCl3, sold under the brand name Lumark among others, is used for radiolabeling other medicines, either as an anti-cancer therapy or for scintigraphy (medical radio-imaging). Its most common side effects are anaemia (low red blood cell counts), thrombocytopenia (low blood platelet counts), leucopenia (low white blood cell counts), lymphopenia (low levels of lymphocytes, a particular type of white blood cell), nausea (feeling sick), vomiting and mild and temporary hair loss. 2) Lutetium (177Lu) oxodotreotide, also known as Lutetium (177Lu) dotatate ("Lutathera"), has been approved for some types of tumors. 3) Lutetium (177Lu) vipivotide tetraxetan ("Pluvicto") has also been approved.
=== Veterinary care === In the United States, more than 15,000 domesticated animals are bitten by snakes each year. Rattlesnake envenomations account for 80% of the deadly incidents. Dogs are most commonly bitten on the front legs and head. Horses generally receive bites on the muzzle, and cattle on their tongues and muzzles. If a domesticated animal is bitten, the hair around the bite should be removed so the wound can be clearly seen. The crotaline Fab antivenom has been shown to be effective in the treatment of canine rattlesnake bites. Symptoms include swelling, slight bleeding, sensitivity, shaking, and anxiety.

== Wound repair == Normal wound repair consists of three different phases: hemostasis and inflammation, proliferation and tissue remodeling. In disturbed wound healing, these stages cannot be completed often resulting in a reduced anatomical and functional outcome. Multiple factors determine the average healing time of the different phases. These factors can be classified into local factors such as infection and ischemia, and systemic factors such as age, stress, Diabetes Mellitus and smoking. In chronic wounds, factors as mentioned above, make it impossible for the tissue to regenerate properly. After injury, the extracellular matrix, and thereby also the heparan sulfate is broken down by different local enzymes, produced by macrophages such as, heparanases, serine proteases and metalloproteinases (MMPs). Heparan sulfate analogues replace the broken heparan sulfate at the wound site and bind to the free heparan sulfate binding sites of the extracellular matrix. Heparan sulfate is slightly negatively charged and so it can bind the positively charged units of the proteins and secure the ECM scaffold. That ensures a supply of the different protein ligands at the wound site.
=== Three dimensional artifacts === Three-dimensional artifacts that have been damaged often require Full backings, but are difficult to apply to three-dimensional textiles. Tailoring techniques such as darts, gathering, similar to original construction techniques can be employed to create shaped backings or supports Occasionally, disassembly is permitted for three-dimensional materials. Shaped forms are also sometimes used as supports. When textiles are used in three-dimensional structures such as covered boxes and upholstered furniture are damaged an overlay can be stitched into lower layers. A less invasive treatment option is passive support. Conditions for use of passive supports: - no major structural damages (tears or holes) - The ground fabric must be intact. - Ideally the textile on its original strainer. - Requires protection against puncture. - Supplementary supports help in limit damages as a result of vibration, shock, and flexing. - The fabric requires additional protection because it is sagging on the strainer. - In the event that the textile has never been removed from its original strainer. The below techniques must be modified to accommodate original lacing: A padded insert provides passive protection. A padded, fabric-covered insert should be created specifically for the object to fill the strainer. Polyester felt or batting can be utilized for a precise fit. A rigid backing (archival-quality rag board of appropriate thickness/rigidity), should be affixed to the insert by thread ties, sewing, or adhesives is used to keep the padded insert secure.
==== Acid pigmentation ==== In 2009 Luigi Garlaschelli, professor of organic chemistry at the University of Pavia, stated that he had made a full-size reproduction of the Shroud of Turin using only medieval technologies. His cloth was woven in the exact same manner and herringbone pattern by yarn type and weight as the Shroud. Garlaschelli placed a linen sheet over a volunteer and then rubbed it with an acidic pigment for the body. The shroud was then aged in an oven before being washed to remove the pigment. He then added blood stains, scorches and water stains to replicate the original. Giulio Fanti, professor of mechanical and thermic measurements at the University of Padua, commented that "the technique itself seems unable to produce an image having the most critical Turin Shroud image characteristics". Garlaschelli noted that the microscopic properties of his reproductions can't be exactly identical to the original, as accelerated and artificial aging lasting 4 hours cannot replicate the natural centuries the Shroud of Turin has gone through. He therefore considered the criticisms of those who claim a need for an absolute similarity as specious. Garlaschelli's reproduction was shown in a 2010 National Geographic documentary. Garlaschelli's technique included the bas-relief approach (described below) but only for the image of the face. The resultant image was visibly similar to the Turin Shroud, though lacking the uniformity and detail of the original.
=== Expansion into Canada === In October 2008, Panera Bread expanded into Canada, opening locations in Richmond Hill, Thornhill, Oakville, and Mississauga in the Greater Toronto Area. A class action lawsuit was filed against the company in February 2008, alleging it failed to disclose material adverse facts about the company's financial well-being, business relationships, and prospects. In February 2011, Panera agreed to pay $5.75 million to shareholders while admitting no wrongdoing, settling the lawsuit. In November 2010, Panera Bread relocated its headquarters to Sunset Hills while vacating its Richmond Heights headquarters and Brentwood, Missouri offices. The company leased additional space for its headquarters in 2013.
=== Murine === Initial therapeutic antibodies were murine analogues (suffix -omab). These antibodies have: a short half-life in vivo (due to immune complex formation), limited penetration into tumour sites and inadequately recruit host effector functions. Chimeric and humanized antibodies have generally replaced them in therapeutic antibody applications. Understanding of proteomics has proven essential in identifying novel tumour targets. Initially, murine antibodies were obtained by hybridoma technology, for which Jerne, Köhler and Milstein received a Nobel prize. However the dissimilarity between murine and human immune systems led to the clinical failure of these antibodies, except in some specific circumstances. Major problems associated with murine antibodies included reduced stimulation of cytotoxicity and the formation of complexes after repeated administration, which resulted in mild allergic reactions and sometimes anaphylactic shock. Hybridoma technology has been replaced by recombinant DNA technology, transgenic mice and phage display.

=== Significance for the origin of life === Nitriles may have played a significant role in chemical evolution on Earth. Experimental studies have demonstrated that hydrogen cyanide can form under a wide range of plausible prebiotic conditions. Possible starting materials include gas mixtures of methane, carbon dioxide, nitrogen, ammonia, and/or hydrogen. Various energy sources, such as electrical discharges or ultraviolet radiation, are likewise conceivable. Under simple conditions, hydrogen cyanide can give rise to numerous additional organic molecules. Hydrogen cyanide and other nitriles, such as cyanoacetylene and dicyan, are considered potential precursors of nucleic bases. Aminonitriles, in turn, are regarded as likely precursors of amino acids and peptides; for example, aminoacetonitrile is a precursor of glycine. An analogous process to the Strecker synthesis is proposed, in which α-aminopropionitrile initially forms from cyanide, acetaldehyde, and ammonia and is subsequently hydrolyzed to alanine.
=== Bay of Bengal === India’s size and position means it has multiple roles in the southern route, both as a destination country and transit point for further destinations. The UNODC has estimated that 54% of the heroin in India is produced domestically, while 45% originates from Afghanistan. India is particularly vulnerable to the southern route due to its western border with Pakistan. Near this border, in the western Indian states of Punjab and Haryana, is where many of the heroin seizures occur. In 2012, 105 kg of drugs were seized, which had been trafficked from Pakistan along rail routes. In 2013 alone, the Indian Narcotics Control Bureau reported seizures totalling 4,609 kg. Data collected through seizures by various authorities has confirmed India as a transit country for Southeast Asia, West Africa and North America. Bangladesh also faces significant problems due to drug trade through the Indian Ocean and India. The country suffers from illicit drug use among its population, such as in Dhaka where there are an estimated 2.5 million people using drugs. India is a large provider of heroin to the Bangladeshi market, and it is trafficked over the western and eastern borders. However, it is unclear whether the heroin originates from Afghanistan or India, as this data has not been sufficiently collected. Both India and Bangladesh are becoming ever more dependent on maritime trade, with these states importing over US$52 million and US$447 million respectively.
Stage 1: Intact skin with non-blanchable redness of a localized area usually over a bony prominence. Darkly pigmented skin may not have visible blanching; its color may differ from the surrounding area. The area differs in characteristics such as thickness and temperature as compared to adjacent tissue. Stage 1 may be difficult to detect in individuals with dark skin tones. May indicate "at risk" persons (a heralding sign of risk). Stage 2: Partial thickness loss of dermis presenting as a shallow open ulcer with a red pink wound bed, without slough. May also present as an intact or open/ruptured serum-filled blister. Presents as a shiny or dry shallow ulcer without slough or bruising. This stage should not be used to describe skin tears, tape burns, perineal dermatitis, maceration or excoriation. Stage 3: Full thickness tissue loss. Subcutaneous fat may be visible but bone, tendon or muscle are not exposed. Slough may be present but does not obscure the depth of tissue loss. May include undermining and tunneling. The depth of a stage 3 pressure ulcer varies by anatomical location. The bridge of the nose, ear, occiput and malleolus do not have (adipose) subcutaneous tissue and stage 3 ulcers can be shallow. In contrast, areas of significant adiposity can develop extremely deep stage 3 pressure ulcers. Bone/tendon is not visible or directly palpable. Stage 4: Full thickness tissue loss with exposed bone, tendon or muscle. Slough or eschar may be present on some parts of the wound bed. Often include undermining and tunneling.
=== Reported end of outbreak: August–September 2022 === On 5 August 2022, the country reported that it had not seen a fever case for seven consecutive days, and that every fever case had recovered. On 8 August, plans were announced to convene the Supreme People's Assembly in September, moving its members out of isolation. A separate review meeting to discuss a "change in direction" in pandemic response was also announced. On 11 August, Kim Jong Un's sister, Kim Yo Jong, stated that her brother had had a fever, implying that he had COVID-19 at one point. She also blamed South Korea for the COVID-19 outbreak in the country, claiming that it sent contaminated anti-Pyongyang leaflets to spread the disease. North Korean media reports indicate that the population have resumed regular activity. On August 25, several sources told NK News that North Korea could possibly allow foreigners into North Korea for the first time since 2020, likely after the 20th National Congress of the Chinese Communist Party later in 2022. In September, however, Kim Jong Un flagged the first rollout of vaccines in November and warned that COVID-19 could reappear in the winter months. The mass vaccination program started in the border areas in late September, making North Korea the last country in the world other than Eritrea to have a mass vaccination program against COVID-19.

Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
Oxidation mainly affects methionine residues and is promoted by dissolved oxygen, trace transition metals, and prolonged exposure to light. Buffer choice and the presence of antioxidants in a formulation can alter the rate appreciably. Because the products differ in mass by only a few units, mass spectrometry is often required to detect them.
Published data on long-term ambient stability are limited, so the question remains open rather than settled. Short excursions during transport are common in practice, and many suppliers use insulated packaging with cold packs. Where stability data are absent, cold-chain handling with temperature logging is the safer approach.
